KASH and SUN proteins

چکیده

KASH and SUN proteins Quick guides at-bat, including which fielders were involved, and so on. At that point, I was amused to see that my friend Kyle was also keeping score, but that he marked every at-bat with a simple and uninformative ‘X’ when each batter was retired; consequently, his whole data sheet on the opposing team was just one string of Xs after another. Although I was feeling the superior data collector, I realized afterward that although I had recorded the game the way I was taught, my data collecting was not up to the task of recording the most significant memory of that game (besides that Koufax pulled off the perfect game). Somewhere around the seventh inning Koufax reached Ball 3 and no strikes, just one errant pitch away from ruining his perfect game. This is the moment when all sound left the stadium and the suspense of the game became indelibly burned into our memories. It turns out that my standard approach to data collecting, including how each batter was put out but not including the count or order of balls and strikes, did not account for the possibility that one might want to have a record of how close Koufax had come to failing. So, Koufax is telling us to use our imagination about where our data might lead, and what might be most truly important.

Novel plant SUN–KASH bridges are involved in RanGAP anchoring and nuclear shape determination

Inner nuclear membrane Sad1/UNC-84 (SUN) proteins interact with outer nuclear membrane (ONM) Klarsicht/ANC-1/Syne homology (KASH) proteins, forming linkers of nucleoskeleton to cytoskeleton conserved from yeast to human and involved in positioning of nuclei and chromosomes. Defects in SUN-KASH bridges are linked to muscular dystrophy, progeria, and cancer. SUN proteins were recently identified ...

Finding the Missing KASH

The nucleus holds and protects the genetic information of a cell. Nuclear positioning is correlated with several important cellular processes, such as cell division, migration and differentiation, and it is essential to the development of organisms ranging from worms to positioning is mediated through a protein complex across the nuclear envelope (NE), formed by the outer nuclear membrane (ONM)...

Identification of unique SUN-interacting nuclear envelope proteins with diverse functions in plants

Although a plethora of nuclear envelope (NE) transmembrane proteins (NETs) have been identified in opisthokonts, plant NETs are largely unknown. The only known NET homologues in plants are Sad1/UNC-84 (SUN) proteins, which bind Klarsicht/ANC-1/Syne-1 homology (KASH) proteins. Therefore, de novo identification of plant NETs is necessary. Based on similarities between opisthokont KASH proteins an...

Interactions between nuclei and the cytoskeleton are mediated by SUN-KASH nuclear-envelope bridges.

The nuclear envelope links the cytoskeleton to structural components of the nucleus. It functions to coordinate nuclear migration and anchorage, organize chromatin, and aid meiotic chromosome pairing. Forces generated by the cytoskeleton are transferred across the nuclear envelope to the nuclear lamina through a nuclear-envelope bridge consisting of SUN (Sad1 and UNC-84) and KASH (Klarsicht, AN...

SUN-1 and ZYG-12, mediators of centrosome-nucleus attachment, are a functional SUN/KASH pair in Caenorhabditis elegans.

Klarsicht/ANC-1/Syne/homology (KASH)/Sad-1/UNC-84 (SUN) protein pairs can act as connectors between cytoplasmic organelles and the nucleoskeleton. Caenorhabditis elegans ZYG-12 and SUN-1 are essential for centrosome-nucleus attachment. Although SUN-1 has a canonical SUN domain, ZYG-12 has a divergent KASH domain. Here, we establish that the ZYG-12 mini KASH domain is functional and, in combinat...

LINC Complexes Form by Binding of Three KASH Peptides to Domain Interfaces of Trimeric SUN Proteins

Linker of nucleoskeleton and cytoskeleton (LINC) complexes span the nuclear envelope and are composed of KASH and SUN proteins residing in the outer and inner nuclear membrane, respectively. LINC formation relies on direct binding of KASH and SUN in the perinuclear space. Thereby, molecular tethers are formed that can transmit forces for chromosome movements, nuclear migration, and anchorage. W...
